As root, or via sudo, just install kernel devel:

sudo yum install kernel-devel

and then run

sudo service vboxdrv  setup

and that should then build the vbox driver and install it.

I am not sure what had done in the past to get  
/usr/src/kernels/2.6.27.12-170.2.5.fc10.x86_64
You or someone had probably booted kernel 2.6.27.12-170.2.5.fc10.x86_64 
and then installed
kernel-devel.
